摘要:透射电镜的样品制备是一个复杂且关键的过程,它直接影响到最终成像的质量和分辨率。透射电镜样品制备的主要目标是获得对电子束透明的薄区,同时保持样品成像的高分辨率和不失真。本报告将对透射电镜样品特征、常见透射电镜试样种类、透射电镜样品的制备方法与设备以及试样保存等方面进行简要介绍。本报告还将简要介绍Fischione相关设备在使用中的部分技巧以及注意事项。
Abstract: Transmission electron microscopy (TEM) specimen preparation is a complex and important process, which directly affects the quality and resolution of TEM images. The purpose of TEM specimen preparation is to get the electron transparent thin regions, and to ensure no artifacts are introduced to the specimen. This report will briefly introduce the features of TEM specimen, types of common TEM specimen, TEM specimen preparation method and instrument as well as the storage of TEM specimen. This report also includes some details and practical skills about how to use the Fischione’s instruments to prepare great TEM specimen.
